St John's was consecrated on 26 June 1846, on land given by J W H Anson, with much of the cost of building provided by the Marshall family.
A Bishop's Instrument, 21 July 1851, assigned a District, from the parish of Manchester. This was confirmed by Order in Council,8 June 1854, (London Gazette, 16 June 1854), and Order in Council, 24 June 1856, (London Gazette, 1 July 1856).
Part of the parish was transferred to St Mark, Gorton, by Order in Council, 26 July 1866, (London Gazette, 27 July 1866), and part to St John Chrysostom, Victoria Park, by Order in Council, 16 May 1878, (London Gazette, 21 May 1878).
A further part was assigned to St Agnes, Birch, by Order in Council, 16 August 1886, (London Gazette, 20 August 1886).
In 1898 the District of St Cyprian, Kirkmanshulme, was formed from St John's parish, by Order in Council, 19 May, (London Gazette, 20 May 1898).
In 1911 another portion was added to St John Chrysostom, by Order in Council, 4 May 1911, (London Gazette, 9 May 1911)
In 1974, by Order in Council, 10 April, (London Gazette, 18 April 1974), the parishes of St John, Longsight, and St Cyprian, Kirkmanshulme, were united as St John with St Cyprian, Longsight, the church of St John becoming the parish church of the united benefice.
